O echip\u0103 de cercet\u0103tori studiaz\u0103 \u00een prezent acest proces \u0219i caut\u0103 modalit\u0103\u021bi de a-l accelera.<\/p>\n<p>Pentru Massimo Vassalli, profesor de bioinginerie la Universitatea din Glasgow, obiectivul este s\u0103 \u00een\u021beleag\u0103 nu doar cum se formeaz\u0103 \u021besuturile vii, ci \u0219i cum pot fi recreate \u00een laborator.<\/p>\n<p>Vassalli este coordonatorul \u0219tiin\u021bific al <a href=\"https:\/\/prism-livingtissues.eu\/\" target=\"_blank\" rel=\"noreferrer noopener\">PRISM-LT<\/a>, un proiect finan\u021bat de UE care se deruleaz\u0103 pe o perioad\u0103 de cinci ani, p\u00e2n\u0103 \u00een 2027. Echipa dezvolt\u0103 o platform\u0103 de bioimprimare 3D pentru crearea de \u021besuturi vii complexe, cu aplica\u021bii de la cercetarea biomedical\u0103 p\u00e2n\u0103 la carnea de cultur\u0103.<\/p>\n<p>La baza proiectului se afl\u0103 un concept care sun\u0103 aproape futurist: materialele vii ob\u021binute prin inginerie, cunoscute sub acronimul ELM (<em>engineered living materials<\/em>).<\/p>\n<p>Este vorba despre materiale realizate par\u021bial sau \u00een \u00eentregime din celule vii \u2013 inclusiv microorganisme precum bacteriile sau ciupercile \u2013 care pot cre\u0219te, pot reac\u021biona \u0219i se pot adapta la mediul \u00eenconjur\u0103tor. Materialele ELM se pot autoorganiza \u0219i autorepara a\u0219a cum materialele statice conven\u021bionale nu o pot face.<\/p>\n<p>\u201eMaterialele vii ob\u021binute prin inginerie pot avea caracteristici suplimentare \u0219i dinamice pe care pur \u0219i simplu nu le putem reproduce \u00een materialele statice tradi\u021bionale\u201d, a declarat Vassalli.<\/p>\n<h3 class=\"wp-block-heading\" id=\"h-formarea-structurilor-din-celule-vii\">Formarea structurilor din celule vii<\/h3>\n<p>Materialele ELM ar putea transforma diverse sectoare, de la s\u0103n\u0103tate p\u00e2n\u0103 la produc\u021bia alimentar\u0103, \u00eens\u0103 transformarea acestui poten\u021bial \u00een realitate presupune rezolvarea unei probleme biologice dificile: cum pot fi imprimate celule vii \u00een structuri complexe f\u0103r\u0103 a le distruge \u0219i f\u0103r\u0103 a pierde controlul asupra modului \u00een care se dezvolt\u0103.<\/p>\n<p>Echipa PRISM-LT abordeaz\u0103 aceast\u0103 problem\u0103 prin crearea de \u021besuturi vii din capsule minuscule care con\u021bin celule vii \u0219i un material de sus\u021binere gelatinos, numit biocerneal\u0103.<\/p>\n<p>\u201e\u00cen loc s\u0103 imprim\u0103m un flux continuu de biocerneal\u0103, lucr\u0103m cu mici unit\u0103\u021bi vii \u00eencapsulate\u201d, a explicat Laura Martinelli, coordonatoarea proiectului PRISM-LT \u0219i directoare general\u0103 la In Society, o organiza\u021bie de cercetare cu sediul la Udine, Italia.<\/p>\n<p>\u201eAceste capsule pot fi pozi\u021bionate cu precizie de un bra\u021b robotic sau pot fi bioimprimate strat cu strat pentru a crea arhitecturi tisulare complexe.\u201d<\/p>\n<p>Metodele conven\u021bionale imprim\u0103 celulele \u00eentr-un flux continuu de material, f\u0103r\u0103 ghidajul biologic pe care \u00eel pot oferi microorganismele vii. Aici, fiecare capsul\u0103 reprezint\u0103 o unitate biologic\u0103 ce con\u021bine at\u00e2t structura de sus\u021binere, c\u00e2t \u0219i microorganismele modificate genetic care contribuie la ghidarea celulelor pe m\u0103sur\u0103 ce se dezvolt\u0103.<\/p>\n<p>Microbii respectivi au fost modifica\u021bi genetic pentru a ac\u021biona ca ghizi biologici. Ei detecteaz\u0103 momentul \u00een care celulele stem \u2013 celule care se pot dezvolta \u00een numeroase tipuri diferite de \u021besut \u2013 \u00eencep s\u0103 se transforme \u0219i reac\u021bioneaz\u0103 prin eliberarea de semnale chimice, numite factori de cre\u0219tere, care ghideaz\u0103 celulele stem c\u0103tre tipul de \u021besut dorit.<\/p>\n<p>Procesul de fabrica\u021bie este rapid, dur\u00e2nd de la c\u00e2teva minute p\u00e2n\u0103 la o or\u0103. Urmeaz\u0103 apoi o etap\u0103 mai lent\u0103: o perioad\u0103 de maturare de aproximativ trei s\u0103pt\u0103m\u00e2ni, \u00een care celulele stem se transform\u0103 \u00een \u021besut osos, adipos sau muscular. Echipa poate produce \u00een prezent aproximativ un centimetru p\u0103trat de \u021besut sub\u021bire \u0219i lucreaz\u0103 la ob\u021binerea unei structuri tridimensionale de un centimetru cub.<\/p>\n<p>Dificultatea este cu at\u00e2t mai mare cu c\u00e2t procesul presupune plasarea \u00een acela\u0219i mediu a unor componente vii care nu coexist\u0103 \u00een mod natural.<\/p>\n<p>\u201eTrebuie s\u0103 cre\u0103m o rela\u021bie simbiotic\u0103 \u00eentre dou\u0103 sisteme care nu au fost concepute s\u0103 convie\u021buiasc\u0103, cum ar fi drojdia \u0219i celulele stem\u201d, a declarat Vassalli. \u201ePrincipala provocare const\u0103 \u00een crearea unor condi\u021bii suficient de bune at\u00e2t pentru drojdie sau bacterii, c\u00e2t \u0219i pentru celulele stem, pe m\u0103sur\u0103 ce acestea se diferen\u021biaz\u0103.\u201d<\/p>\n<p>Aceast\u0103 curiozitate legat\u0103 de interac\u021biunile biologice a stat la baza proiectului. \u201ePunctul de plecare al cercet\u0103rii noastre a fost curiozitatea fa\u021b\u0103 de aceast\u0103 interac\u021biune\u201d, a spus Vassalli. \u201e\u00cen esen\u021b\u0103, evolu\u021bia a urmat acela\u0219i proces. Organismele unicelulare au interac\u021bionat \u0219i au evoluat, d\u00e2nd na\u0219tere naturii a\u0219a cum o cunoa\u0219tem ast\u0103zi.\u201d<\/p>\n<h3 class=\"wp-block-heading\" id=\"h-de-la-cercetarea-maduvei-osoase-la-carnea-din-farfurie\">De la cercetarea m\u0103duvei osoase la carnea din farfurie<\/h3>\n<p>Cercet\u0103torii \u00eencearc\u0103 s\u0103 recreeze dou\u0103&nbsp;tipuri specifice de \u021besuturi. Primul este interfa\u021ba dintre \u021besutul osos \u0219i cel adipos, prezent\u0103 \u00een m\u0103duva osoas\u0103, pentru cercetarea biomedical\u0103.<\/p>\n<p>Al doilea este reprezentat de structuri alc\u0103tuite din mu\u0219chi \u0219i gr\u0103sime, care reproduc stria\u021biile de gr\u0103sime ce dau c\u0103rnii reale textura \u0219i gustul specifice \u2013 o calitate care a lipsit mult timp din carnea de cultur\u0103, sau carnea produs\u0103 \u00een laborator.<\/p>\n<p>Platforma va fi utilizat\u0103, de asemenea, pentru a crea modele tisulare miniaturale care imit\u0103 structura organelor umane \u0219i care ar putea fi folosite pentru testarea medicamentelor \u0219i pentru sprijinirea medicinei personalizate.<\/p>\n<p>\u201eProiectul \u00ee\u0219i propune s\u0103 ofere o platform\u0103 care poate crea prin inginerie \u021besuturi diferite, cu utiliz\u0103ri foarte diverse, dar plec\u00e2nd de la acelea\u0219i principii de baz\u0103\u201d, a declarat Martinelli.<\/p>\n<p>\u00cen domeniul s\u0103n\u0103t\u0103\u021bii, accentul se pune pe realizarea de modele 3D ale m\u0103duvei osoase pentru studierea medicamentelor destinate unor afec\u021biuni care afecteaz\u0103 \u00een mod specific m\u0103duva osoas\u0103, cum ar fi leucemia.<\/p>\n<p>\u00cen ceea ce prive\u0219te produsele alimentare, ob\u021binerea unei distribu\u021bii adecvate a gr\u0103simii este foarte important\u0103 pentru acceptarea lor de c\u0103tre consumatori. \u201eDatorit\u0103 tehnologiei noastre de bioimprimare, putem ob\u021bine textura potrivit\u0103 pentru alternativele la carne, ceea ce ar putea facilita introducerea acestora pe pia\u021b\u0103\u201d, a declarat Martinelli.<\/p>\n<p>Mai este \u00eens\u0103 nevoie de timp pentru ca aceast\u0103 tehnologie s\u0103 ajung\u0103 la publicul larg. \u201eSuntem \u00eenc\u0103 departe de aplica\u021biile practice\u201d, a declarat ea. \u201eNe concentr\u0103m pe principiile \u0219i mecanismele de baz\u0103, pentru a vedea ce func\u021bioneaz\u0103 \u0219i ce nu. Introducerea materialelor vii ob\u021binute prin inginerie \u00een medicin\u0103 sau \u00een produc\u021bia alimentar\u0103 va necesita, totodat\u0103, o nou\u0103 perspectiv\u0103 \u00een materie de reglementare.<\/p>\n<p>Deoarece materialele ELM combin\u0103 celule vii \u0219i, \u00een unele cazuri, microorganisme modificate genetic, nu se potrivesc perfect cu reglement\u0103rile existente \u2013 concepute pentru medicamente conven\u021bionale sau produse alimentare standard, nu pentru materiale care sunt, practic, vii.<\/p>\n<p>\u00cen colaborare cu Consiliul European pentru Inovare, echipa poart\u0103 deja discu\u021bii cu autorit\u0103\u021bile de reglementare, inclusiv cu Agen\u021bia European\u0103 pentru Medicamente, pentru a analiza ce norme \u0219i autoriza\u021bii ar putea fi necesare \u00een viitor pentru aceste materiale.<\/p>\n<p>\u201eTrebuie s\u0103 privim aceast\u0103 tehnologie dintr-o nou\u0103 perspectiv\u0103\u201d, a declarat Martinelli. \u201eAceast\u0103 colaborare ne ajut\u0103 s\u0103 identific\u0103m o cale pentru utilizarea materialelor ELM.\u201d<\/p>\n<p>Vassalli a ad\u0103ugat c\u0103, utilizate pe scar\u0103 larg\u0103, materialele ELM ar putea fi \u201eextrem de puternice\u201d. \u201eC\u00e2nd am demarat proiectul, ne-am pus dou\u0103 \u00eentreb\u0103ri principale: se poate realiza \u0219i, mai ales, poate fi aplicat la scar\u0103 larg\u0103?
